What Mitochondria Do
Mitochondria are the organelles responsible for producing most of a cell's usable energy (ATP) through oxidative phosphorylation, and they're present in nearly every cell in the body in numbers proportional to that tissue's energy demand — muscle and heart cells, for instance, are packed with them. Beyond energy production, mitochondria play roles in calcium signalling, cell death regulation, and generating reactive oxygen species that, at controlled levels, act as signalling molecules rather than purely as damage.
Why They're Central to Ageing Research
Mitochondrial dysfunction is formally recognised as one of the hallmarks of ageing in the influential biogerontology literature, alongside things like genomic instability and cellular senescence. With age, mitochondrial DNA accumulates mutations, mitochondrial number and efficiency in tissues like skeletal muscle tend to decline, and the quality-control process that clears damaged mitochondria (mitophagy) becomes less efficient — a combination linked to reduced energy capacity and increased oxidative stress in ageing tissue.
What Actually Moves Mitochondrial Function
Of the interventions studied, exercise — both aerobic training and resistance training — has the most consistent, well-replicated evidence for improving mitochondrial density and function in skeletal muscle, in some studies producing changes comparable to what's seen with certain pharmacological interventions in animal models. Caloric restriction and time-restricted eating also show mitochondrial benefits in both animal and some human studies, plausibly linked to the same pathways activated by fasting.
Where Supplement Claims Get Ahead of the Science
A long list of supplements (CoQ10, PQQ, NAD+ precursors, and others) are marketed on mitochondrial-support claims, and some have genuine cell-culture or animal data behind specific mechanisms — but human trial evidence for measurable clinical benefit from supplementation alone is generally much weaker and more mixed than the marketing suggests. Exercise and sleep remain the best-supported levers; anyone considering supplementation for a specific concern should discuss it with a licensed physician, particularly given potential interactions with existing medications.
